植物生理學實驗以配合正課教學內容為主軸,內容可分成幾大主題,每個主題包含多個實驗課程;I、水份輸送:利用簡單的實驗方法及儀器輔助測定植物中水勢大小及蒸散作用速率,了解水份在植物體中吸收、運送、蒸散的原理及意義;II、營養元素吸收與功能:觀察植物生長發育對特定營養元素的需求及缺乏時所引發的病徵,並藉由根瘤菌之接種了解植物界的共生現象,更進一步學習定量leghemoglobin蛋白質及利用氣相層析儀分析根瘤中固氮酵素的作用活性,比較分析外在元素對共生作用及固N2活性的影響;III、光合作用:學習利用離心法萃取單離葉綠體進行光合作用速率的測定;利用分光光度計分析多種色素的吸收光譜,比較光合色素和花青素的吸收光譜,並了解其不同生理功能的意義;IV、大分子抽取及分析:學生將練習抽取植物體中之醣類、酵素、蛋白質等有機分子,除初步定量及定性分析,亦將學習蛋白質電泳技術、利用電腦計算及分析結果。V、植物荷爾蒙功能檢定:探討多種植物荷爾蒙的專一性及交互性生理功能,並學習利用無菌培養技術以進行植物分化全能性之實驗;VI、逆境生理:研究環境逆境對植物所引發的抵抗機制,並利用生物染色法檢試細胞活性及以離子測定儀測量細胞膜滲漏情形。
