一、課程簡介:
本實驗課程主要目標,在介紹並實際操作微生物研究之相關基本方法。本課程為一學分,每週三小時。實驗內容包括,文獻之查詢及排序,培養基之製備,純粹培養之分離技術,顯微鏡之使用,細菌之染色,細菌生長之測定,生理及生化特性之分析,抗生素敏感性之測定,突變株之篩選,核酸之純化,PCR反應及電泳之分析,免疫偵測方法,真菌產孢之誘導及觀察,以及噬菌體之培養分離與計數等。學期成績主要由實驗報告及期末考試為依據。
The goal of this laboratory course is to introduce students some basic methods in microbiological research. This course is one credit hours and three hours every week. The grade will be determined by the lab reports and final examination.
Contents:
1. Class organization and library orientation
2. Medium preparation
3. Techniques for isolation of pure cultures
4. Microscopic examination
5. Staining of bacteria – simple stain, Gram stain, and negative stain
6. Staining of bacteria – the endospore, flagella stain, and capsule stain
7. Measurement of bacterial growth – standard plate count and turbidometric assay
8. Effect of osmotic pressure and temperature on the growth and morphology of bacteria
9. Biochemical tests – starch hydrolysis, oxidase production, oxidase production, protein digestion, formation of fluorescent pigment
10. Antibiotic sensitivity testing
11. Isolation of spontaneous mutant
12. Isolation of genomic DNA from bacteria
13. PCR analysis and agarose gel electrophoresis
14. Immunodetection - ELISA and immunofluorescence staining
15. Sporulation of fungi
16. Cultivation and enumeration of bacteriophages
Grade rating: 50% report, 45% examination, 5% lab performance
