課程概述

Biomedical optics has been increasingly applied to the screening, surveillance and diagnosis of diseases, as well as the non-invasive monitoring of physiological status of tissue/organs in vivo. The interaction between light and biological tissue involves mainly scattering and absorption, which can be modeled analytically with the framework of the Radiative Transport Equation (RTE) or numerically with the Monte Carlo method. With sufficient understanding and appropriate modeling of light-tissue interactions, the clinically relevant and useful information can be extracted from optical signals measured from tissue. The following techniques will be introduced:
1. Diffuse reflectance spectroscopy (DRS) and imaging
2. Near-infrared spectroscopy (NIRS) and tomography
3. Tissue oximetry using DRS or NIRS
4. Fluorescence spectroscopy
5. Single (few) scattering techniques
6. Raman scattering spectroscopy
Other optical techniques related to the non-invasive diagnosis or monitoring of physiological conditions will be reviewed by the students (topics chosen by the students). Note that techniques using only ballistic photons to provide signals (e.g. confocal microscopy and non-linear microscopy) are NOT covered in this course. 

課程目標
To obtain the fundamental knowledge and skills for the analysis and interpretation of data measured from tissue, the following steps will be employed:
1. The principle of light propagation in soft tissue will be introduced.
2. Students will learn the practical use of the Monte Carlo method to numerically simulate light propagation in soft tissues.
3. The students will review the literature on selected optical techniques that can be used to aid the non-invasive diagnosis of disease or monitoring of physiological status.
4. Students will get hands-on experience with Monte Carlo simulations or the RTE framework by working on a term project with a topic related to optical diagnostic techniques. 
課程要求
1. Homework (individual)
Each student will develop a computer program using the Monte Carlo method to simulate the propagation of photons in biological tissues. Seven sets of computer assignments will be given to gradually guide the students through the model development process. Basic programming ability or acquaintance with Matlab is required.
Alternatively, students can choose Track 2 in which they submit a progress report (with the same due date as the homework) instead of the computer homework.
2. Literature review (individual)
To gain a more in-depth understanding of a selected technique, each student is required to pick a specific topic, study related literature, and make an oral presentation to introduce the selected topic. In addition, students are expected to participate in discussion.
3. Term project
Students will apply the simulation method they have learned in class to improve the data interpretation/analysis or experimental design, or to address a specific question related to optical diagnosis/monitoring of disease/physiology.
Students who choose Track 2 are required to prepare a draft for submitting to a conference or a journal.
